After designing and building leading synthetic antibody libraries with several partners we have produced our own groundbreaking SuperHuman antibody library. It was designed as a consequence of seven years of natural repertoire analysis and synthetic repertoire design. In previous natural libraries, it was observed that expanded memory clones introduced redundancy that limits library diversity to be at least 100-fold less than the number of transformants. Synthetic libraries avoid generating expanded clones, but produce large populations of antibodies with sequences with non-functional diversity combinations: between 90-99% of antibodies in synthetic libraries can be assumed to have folding or stability defects as a consequence of unnatural diversity combinations. In the SuperHuman library we choose therapeutically-optimal frameworks and load them with naturally selected diversity in a mathematically optimal manner. The resulting library is then subjected to a thermal tempering process to produce an ultra-diverse library where every molecule is functionally folding, well expressing, and very unique and distinct from other library members. On top of our analysis platforms we also offer Bioengineering services.
